Abstract: Aus UV-spektroskopischen Vergleichen mit zwei durch H2O2-Oxydation von Phalloidin erhaltenen Sulfoxiden (1b und 1c) und mit Skatyl-(2)-äthyl-sulfoxid (2b) folgt die Sulfoxid-Natur des Amanins (7e). Auch die Amanitine (7a, 7c, 7d) sind Sulfoxide. Das UV-Spektrum des Methyl-α-amanitins (7b) stimmt mit demjenigen von synthetisch erhaltenem [6-Methoxy-skatyl-(2)]-äthyl-sulfoxid (5b) überein.

  • ** Alpha-amanitin (AA), beta-amanitin (BA), gamma-amanitin (GA), epsilon-amanitin, amaninamide , amanin, and amanullin are some of the members of amatoxins group (Lindell et al., 1970). AA is also the first purified toxin and it can easily be obtained with high purity today (Faulstich et al., 1973; Kaya et al., 2012b), not only AA but also BA and GA is purified highly (Kaya et al., 2012cKaya et al., , 2013c) The protein synthesis stops in the cell when RNA polymerase II is blocked. AA, after oral intake, clinical toxicity is observed in liver and kidneys.

  • ** To demonstrate the proposed methodology, we chose to analyze samples containing mushroom toxins of the Amanita family [12]. These toxins have been extensively studied during the past 70 years, and the mushrooms are known to produce more than 20 different cyclic peptide toxins, which can be grouped into amatoxins, phallotoxins and virotoxins, where a-amanitin and bamanitin are regarded as the most potent [12] [13] [14] [15] [16] [17] [18] [19] [20] [21] [22] [23]. The Amanita genus includes over 745 species and varieties, but only 9 are known to produce amatoxins [24].

  • ** To determine the quality of the sample preparation, cell suspensions obtained by collagenase/dispase digestion were stained with phalloidin-FITC . Phalloidin binds to filamentous but not monomeric actin [Govindan et al., 1972] and all actin variants have been shown to bind phalloidin [Faulstich et al., 1988].
